Mass isotopomer analysis is an important technique to measure the production and flow of metabolites in living cells, tissues, and organisms. This technique depends on accurate quantifications of different mass isotopomers using mass spectrometry. Constructing calibration curves using standard samples is the most universal approach to convert raw mass spectrometry measurements into quantitative distributions of mass isotopomers. Calibration curve approach has been, however, of very limited use in comprehensive analyses of biological systems, mainly suffering from the lack of extensive range of standard samples with accurately known isotopic enrichment. Here, we present a biological method capable of synthesizing specifically labeled amino acids. These amino acids have well‐determined and estimable mass isotopomer distributions and thus can serve as standard samples. In this method, the bacterium strain Methylobacterium salsuginis sp. nov. was cultivated with partially 13C‐labeled methanol as the only carbon source to produce 13C‐enriched compounds. We show that the mass isotopomer distributions of the various biosynthesized amino acids are well determined and can be reasonably estimated based on proposed binomial approximation if the labeling state of the biomass reached an isotopic steady state. The interference of intramolecular inhomogeneity of 13C isotope abundances caused by biological isotope fractionation was eliminated by estimating average 13C isotope abundance. Further, the predictions are tested experimentally by mass spectrometry (MS) spectra of the labeled glycine, alanine, and aspartic acid. Most of the error in mass spectrometry measurements was less than 0.74 mol% in the test case, significantly reduced as compared with uncalibrated results, and this error is expected to be less than 0.4 mol% in real experiment as revealed by theoretical analysis. Rotation ambiguity (RA) in multivariate curve resolution (MCR) is an undesirable case, when the physicochemical constraints are not sufficiently strong to provide a unique resolution of the data matrix of the mixtures into spectra and concentration profiles of individual chemical components. RA is often met in MCR of overlapped chromatographic peaks, kinetic and equilibrium data, and fluorescence two‐dimensional spectra. In case of RA, a single candidate solution has little practical value. So, the whole set of feasible solutions should be characterized somehow. It is a quite intricate task in a general case. In the present paper, a method was proposed to estimate RA with charged particle swarm optimization (cPSO), a population‐based algorithm. The criteria for updating the particles were modified, so that the swarm converged to the steady state, which spanned the set of feasible solutions. The performance of cPSO‐MCR was demonstrated on test functions, simulated datasets, and real‐world data. Good accordance of the cPSO‐MCR results with the analytical solutions (Borgen plots) was observed. cPSO‐MCR was also shown to be capable of estimating the strength of the constraints and of revealing RA in noisy data. As compared with analytical methods, cPSO‐MCR is simpler to implement, expands to more than three chemical compounds, is immune to noise, and can be easily adapted to virtually all types of constraints and objective functions (constraint based or residue based). cPSO‐MCR also provides natural visual information about the level of RA in spectra and concentration profiles, similar to the methods of two extreme solutions (e.g., MCR‐BANDS). The problem of finding the plane supporting curve (brachistochrone) along which a heavy particle released from rest at the given starting point slides with dry (Coulomb) friction and reaches the given destination point in least time, is stated as a variational isoperimetrical problem. The finite parametric equations of the extremal curve are obtained. This curve is the sought-for brachistochrone if the solution of the problem exists. Several numerical examples are given.  相似文献

In order to study the conceptions, and their evolutions, of the tangent line to a curve an updating workshop which took place in México was designed for upper secondary school teachers. This workshop was planned using the methodology of cooperative learning, scientific debate and auto reflection (ACODESA) and the conception-knowing-concept model (cK¢) developed mainly by Balacheff. In order to initiate the conceptions reorganization, an initial activity was made in the graphic frame; it seems optimal to start the debates without leading participants to failure. The mathematical core of the workshop was formed by an algebraic method to find tangents to algebraic curves which is close to Descartes’ method. The ACODESA methodology allowed some intense debates mainly concerning the local character of a tangent, generating the teachers’ cognitive unbalance which is a starting point for the refinement or transformation of their conceptions. On the other hand, the cK¢ model allows to understand the conceptions of participants and to analyze the evolution of their knowings.  相似文献

For an accurate simulation of forming processes, it is of paramount importance to model the different lubrication regimes that can develop at the contact interface. These might vary from zone to zone of the forming piece, and from one regime to another, resulting in forces of different nature and magnitude.
